Arxula – Scientific FeaturesThe Arxula expression platform is originating from the research of Prof. Dr. G. Kunze, IPK Gatersleben. Basic proof-of-principle for recombinant protein production has been achieved by this group on enzymes and therapeutics, such as a scFv antibody fragment and Interleukin-6 (IL-6). Especially with the example of IL-6, Arxula has proven its general capacity to express difficult proteins in correct form and at significiant productivities. Despite two Proline residues present at position +2 and +3 of the mature IL-6 Molecule, this protein is secreted in the correctly processed form. Comparative expression with the Xplor Vector System has revealed that other yeast show misprocessing from the same precursor. All elements necessary for high level gene expression – vectors, marker, promoter and strains – have been developed and established as reliable tools for commercial projects. The Arxula technology is further developed in close collaboration between IPK and ARTES and focuses now on aspects such as fermentation, whole-cell biocatalysis, secretion and expression optimization. The Arxula technology is exclusively marketed and licensed by ARTES. This dimorphic, non-pathogenic yeast combines the best characteristics of two worlds: yeast and fungi. It is able to assimilate many compounds as only source for carbon and nitrogen. Osmotolerance and thermotolerance as well as an exceptional growth and secretion behaviour make Arxula an attractive expression system. Low- and high-copy co-expression of genes offer a unique tool for the targeted gene expression, for production of proteins, for introduction of new metabolic pathways and for coexpression of bottleneck genes. The most recent success in protein expression from Arxula has been achieved with a single chain antibody at a level of 200 mg/L in shake culture. Arxula – Track Record
